Acquiring TEM Images for Calibration & MeasurementThe TEM magnification was calibrated at 120 V by capturing images with the Gatan CCD camera using Digital Micrograph. Images were saved as TIFF files. The diffraction grating should be handled carefully so as to not distort the grating. To reserve the grating, start at low magnification with one grid square filling the screen, increase the illumination to near max but not crossover, then reduce the illumination. Now go to the desired magnification. When moving to a new grid area, reduce the magnification and illumination, and repeat this procedure. To focus on the grating at low magnifications, use the wobbler button on the left side of the scope and the largest objective aperture. A larger amplification wobble makes it easier to focus at the lowest magnification (directions on how to adjust the amplification of the wobbler are in the Philips user manual). When the image is in focus, the two images from the wobbler will be aligned. |
