Notes:
After piece of biological tissue or other material is embedded into some resin - you need to obtain sections thin enough to be penetrated by the electron beam. For biological structures it is 50-70 nm thick sections.
In our facility we have an excellent instrument for that purpose - state of the art Ultracut S. It has an excellent system of block illumination that provides very easy alignment of the block relatively to the knife edge. Knives used for that purpose - are diamond knives.
